You are eligible to write National Eligibility cum Entrance Test after passing class 12th board examination. Further, your aggregate percentage in physics, chemistry and biology should be above 50%.

Graduation means successfully completing at least three years course after 12th. Even though diploma is three years, it's actually after 10th. In simple English it will mean completion of college after 12th.

ISRO required specialists in different fields. You imagine rockets and satellites. Whatever goes into them and whatever enables launch. Varied fields means varied specialists. Chemical, materials, physics, etc.

I think you cannot change subjects in 12th. You have to do it in 11th only. But yes, you need to have PCB if you are thinking of pursuing career in biotechnology engineering. Best wishes to you for this!
