As per the UGC Norms and AICTE guidelines, None a university in India can offers Engineering & Medical Course from distance mode. Doing this is the violation of policy. Hence, go for regular mode.

Admission is through counselling based on rank obtained in Uttar Pradesh State Entrance Examination Lateral Entry (UPTU SEE).
Other eligibility criteria:
Candidate must have passed 3-years Engineering Diploma or passed B.Sc and class 12th with Mathematics and obtained not less than 45% marks in aggregate (40% in for SC/ST).

Yes, you are eligible for getting admission in MIT WPU Pune as your Diploma aggregate is good enough to get admission there. They will provide you the best knowledge and also the best placement. So you can try to get admission.

Yes, you can go for Ph.D directly. As there is was a programme launched sometime before according to which for doing Ph.D, it is not mandatory to complete your Master's degree first. You can directly go for Ph.D just after your graduation. So, you can do Ph.D. after completing your B.Tech in biotechnology.
