No you can not get admission in Punjab Engineering College on the basis of Engineering Diploma. Diploma holders are eligible to appear for IIT JEE, but they will be considered similar to 12th passed candidates. They will not be given admission in the second year of engineering in IITs, Diploma holders have to appear for JEE Main qualifying which they will be eligible to appear for JEE Advanced exam which will be including two papers where both the papers are compulsory. To appear in JEE Advanced a Diploma holder can apply in JEE Main 2021. But their rank & score in JEE Main will not be considered for admission in NITs & CFTIs.

Technically Yes, A candidate can appear in all the 4 sessions of JEE Main. The eligibility criteria of all the papers is the same. However if dates and timings of Paper-1, Paper-2A and Paper-2B are not overlapping than these sessions can be attempted easily.

B.E. / B.Tech courses can be pursue after the Engineering Diploma courses. So on the basis of Health Inspector Diploma you can not pursue the B.E. /B.Tech courses. However if you have completed the 10+2 with PCM than can be register for AMIE under Technician Member.
