Diploma holders are eligible to appear in JEE (Advanced), therefore, they can apply to appear in JEE Main but rank and score of such students for JEE (Main) is not declared as diploma holders cannot apply for admission in NITs and CFTIs through JEE (Main) channel.
You have to clear JEE Mains followed by JEE Advanced, provided you satisfy the eligibility criteria of JEE.
To get admission in IITs, you need to have at least 75% in class 12th. For general category, it is 75% and for reserved categories, it is 65%. The lower grades will not make you eligible to sit for IIT-JEE Advanced.

Yes, diploma is considered as equivalent of 12th and hence you are eligible to get admission in IIT.
Currently, there is no provision of lateral entry into IIT or NIT after Diploma in engineering.
NIT or IIT don't have B. Tech through the lateral entry you can give exam for entry in 4-year engineering, there are other good colleges like DTU and Thapar which provides lateral entry.
Just check if you are getting admission in a decent institute having good placement record, known for academics. If you are not getting any good college than you can give a try for IIT.
Currently there is no provision of lateral entry admission in IITs and NITs for Diploma engineers.

No one gets into IIT by JEE Mains. You need to qualify for JEE Advance and score high in that to get into IITs.
IITs follow only JEE marks, no other such schemes are there for admission.
