No, you will not lose your one attempt if you do not appear for JEE Mains just after 12th. You can appear for the exam in any of the three consecutive years after passing your 12th board exams. So, if you do not feel prepared to take the exam in your first year after 12th, you can take a year off to prepare and then appear for the exam in your second or third year.
