It depends on the university in which you are planning to apply. But you need to have knowledge of your Bachelor's degree and chemistry. Sometimes, questions from chemistry do come. If you are planning for Delhi University then read your Bachelor's degree syllabus, it's more than enough. For Indraprastha University, study chemistry and Bachelor's degree portion.

No, you cannot open a wholesale business for medicine without licence. Drug licence (DL) is required for selling medicine. First, get the license by doing pharmacy courses like D.Pharma or B.Pharma and then you need to apply for wholesale business.
