You can pursue Ph.D after completing M.Sc and it is the normal route. You don't have to pursue M.Tech to pursue Ph.D, unless you want to do it.

Getting a Government Job after the age of 35 is a little difficult because the age limit in most of exams is less than 30 years (Bank PO, SSC CGL, Railway Exams etc.)
If you belong to the reserved category then you can check the age relaxation & apply for the exam.
Well you can also qualify the exam for Lectureship, by preparing for the CSIR NET Exam.
CSIR NET Exam is the national level eligibility test for Science students to become Research Fellow or Assistant Professor. But the age limit for Research Fellow is 28 years.
So, you are eligible for Lectureship. You can get a job in Government colleges & universities

After doing M.Sc. Microbiology, one can join R&D department of any good Biotech company.
You can also join research programmes after clearing UGC or CSIR NET exam & further go for PhD.
