It is not compulsory for you to give an entrance exam for admission to the BTech Nanotechnology course. There are around 21 colleges in India that offer the course, out of which more than 10 colleges give admission on the basis of the Class 12 board exam marks. So, you should work hard and prepare well for the board exams to score excellent marks.
Having said that, it is better to prepare for the entrance test and keep both options open. Best of luck.

To get admission to your dream college for the BTech Nanotechnology course, you should start preparing for the entrance exam at least 8 to 10 months before the tentative exam date. It gives you enough scope to cover the maximum syllabus sections, practice previous year question papers, take mock tests, and plan a rigorous revision schedule in the last 1-2 months.
The time may also differ on the basis of the exam you're planning to appear for. In the case of a national-level exam like JEE Main, you need extra practice and preparation owing to its competitiveness and difficulty.

Well, yes. If you belong to any of the reserved categories, including SC/ST/OBC/PwD, you usually get some sort of marks relaxation at the time of BTech Nanotechnology admission. The general relaxation is 5% at a majority of the colleges. For example, if the eligibility requirement is 50% marks for the general category, the reserved category students need 45% marks, with a similar approach in the entrance exam cutoffs.
Res, you must go to the college websites where you plan to pursue the course to have better clarity.
