Yes, Your bother will be eligible to get a seat through JEE Main if he clears his Class 12th supplementary exams. Clearing of Class 12 is a mandatory requirement. Validity of supplementary exams will depend upon clearing the subjects required.
For admission to NITs, IIITs, and GFTIs, he must have obtained at least 75% marks in Class 12 or must be within the top 20 percentile of his board (65% for SC/ST). I wrote both JEE and neet. so with my experience, Let me give you this details:
NEET Eligibility
The age of the candidate must be 17 years as of December 31 of the admission year. Candidates must have completed 10+2 with Physics, Chemistry, Biology/Biotechnology, and English as core subjects, securing 50% marks for general, 40% for OBC/SC/ST, and 45% for PwD candidates.
CUET Eligibility
Age has not been declared. The minimum qualification required for the students should be 10+2 or equivalent from any recognised board. The eligibility for the particular course depends on the university and their requirements.

The candidate must at least score a minimum of 50% marks in their examination. NIT Rourkela offers admission after considering the 12th exam results, JEE Main score. A merit list shall be made category-wise, listing the All India Rank (AIR) of candidates which is used for the Counselling process.
To get admission in NITs, candidates must have a good percentile to get their desired institution and specialization. Generally, candidates should having a percentile between 85 to 95 for getting NIT seats.

70% of admissions to Amrita Vishwa Vidyapeetham Chennai BTech courses are granted based on AEEE scores. As per the university's official website, the schedule for the Phase 1 exam has been released.
